Going to need a bit more info fella - afaik you can spec the Macan from a 340mm all the way up to 390mm rotor's...
Then what size alloys?

Well stock Cupra R brakes are 343mm, and fit comfortably behind the factory fit 19's - so you should be fine? Performance Pack brakes are 370mm!
 
340mm and the cupra borbet 19s which I believe are et45.
If your alloys are the factory OEM alloys (part number 5F0601025K, manufactured by Borbet for Seat) that were available on the mk3 Leon Cupra like the ones below, they’re 8Jx19” with an offset of ET50 rather than ET45. An offset of ET45 is usually found on aftermarket alloys, so if yours are ET45 then it’s possible they may be replicas; if you remove one you can check the spec details, part number, manufacturer name etc. that will be stamped into the rear of the spokes.

Based on the information provided at the link below, when the 340mm macan calipers are fitted to the VW equivalent car to the Leon Cupra (mk7 Golf R with factory 19” VW Pretoria alloys 8Jx19” ET50, also manufactured by Borbet), it states that 10mm spacers are required. Maybe they’re just being cautious if @TripleBob has said they’ll fit comfortably behind factory 19” alloys.

 
Not all the mk3 cupra alloys are et50 the carbon edition wheels are et45 not sure about cupra r wheels.

I would agree and think spacers are required.

One of the reasons/benefits to the common single piston sliding caliper is they are much more compact/thinner allowing larger offset alloys as you don't have pistons on the outside of the disc.
Looking at this image the OE sliding piston already looks 'close' so i'm sure a fixed 4x piston caliper would need wheel spacers to fit.

Also note the 'performance pack' Cupra uses a different knuckle (Same as TTRS/RS3) to the standard 340mm cupra - so maybe the caliper mounting lugs are also set back compared to the wheel mounting surface? The Brembo caliper mounts behind the lugs vs in front like in most sliding calipers.
